Human embryonic stem cells are able to differentiate in culture and produce numerous somatic cell types.We call the process marker succession.The first problem that presents itself is that hES cells, which give rise to so many somatic cell types, also produce teratomas.In order to free any derived somatic cell type from residual hES cells, it will be necessary to clonally isolate the cell type and expand it to mass culture prior to using it for therapeutic replacement.One should not lose sight of the fact that cultured embryonic stem cells are not embryos.The polarity and the gradients that are important in controlling embryogenesis are absent during the differentiation of cultured hES cells.Optimization of culture conditions improves the proliferation, but not sufficiently to permit their clonal isolation.These cells were easily cloned and grown to mass culture.But for prospective therapeutic purposes, it will be preferable to use other genes capable of improving proliferation without perturbing differentiated function.Our current research is devoted to finding the best solution to this
